The build of the game shown at E3 2004 used Grant Kirkhope's voice samples of Donkey Kong. In the final game he was voiced by Takashi Nagasako.

During the E3 2004 demo of the game, DK shouted a basic howl after defeating a boss, rather than the one heard in the final game which fluctuates in tone.

In the E3 2004 build of the game, instead of fighting Dread Kong, the player fought against an unnamed clone of Donkey Kong that had grey fur, no tie, and glowing red pupils.

To promote the game, Nintendo of America sent 50 employees dressed in ape costumes to the 20th annual Los Angeles Marathon in 2005. Some of those runners wore Jungle Beat t-shirts, sweatbands, and carried DK Bongo controllers.

Donkey Kong Jungle Beat was the first game to receive an E10+ rating from the Entertainment Software Ratings Board following the rating's introduction two weeks before the game's North American release in March of 2005.
